Figure 6. Proteinase K digestion studies conducted as part of study of propagation of classical scrapie prions. K222-Tg516 and Q222-Tg501 homozygous mice were inoculated with classical scrapie. A) Proteinase K–resistant prion protein (PrPres) sensitivity in the brains of Q222-Tg501 and K222-Tg516 mice initially inoculated with F10 scrapie isolate and subsequently reinoculated into both the original model and its counterpart. B) PrPres sensitivity in the brains of Q222-Tg501 and K222-Tg516 mice initially inoculated with CP060146 scrapie isolate and subsequently reinoculated into both the original model and its counterpart. In both cases, proteinase K concentrations of 500, 50, 1 and 0.1 µg/mL were tested. Western blot visualizations were done using the Sha31 monoclonal antibody. Molecular weight markers are indicated on the right side of each band.
